A model for the study of skeletal anomalies in rat fetuses BABT
Effting,Cristiane; Paula,Daniel Jesus de; Nunes Junior,Guilhermino Pereira.
The aim of this study was to validate a model of skeletal anomalies in rat fetuses by the administration of ketoconazole (80 mg/kg) to pregnant rats during organogenesis. Bones of the head, trunk and anterior and posterior limbs were examined for detection of anomalies. Statistical differences regarding the number of fetuses and postimplantation resorptions, and fetal and placental weight were significant. The frequency of skeletal anomalies in the head, trunk, and anterior and posterior limbs in the ketoconazole-treated group were also significant when compared to the control group. It could be concluded that the model suggested was valid for study of skeletal anomalies and abnormal bones development in rat fetuse, in spite of the loss of fetuses due to...

Acetaldehyde induced developmental and genetic damage in Drosophila melanogaster JBAG
Palermo,A.M; Mudry,M.D.
Acetaldehyde (AAld) is a ubiquitous compound in the environment. Exposure may occur in its manufacture or use, in the consumption of alcoholic beverages and through cigarette smoke. It causes distant target effects, including DNA damage, glutathione depletion, enzyme inactivation and cell death. In the present work, its potential teratogenic, mutagenic and recombinagenic effects in somatic cells of D. melanogaster were analyzed. The white/white+ (w/w+) eye mosaic test (Somatic Mutation And Recombination Test, SMART) was applied by mating w/w females with y/Y males. Larvae of 48 ± 4 h were transferred to media with 0.01 %, 0.02 %, 0.05 % and 0.10 % AAld for 12 hs and then moved to regular media until pupation. The emerging w/w+ females were evaluated for...

EFFECTS OF LOW-DOSE PHENYTOIN ADMINISTRD TO RATS FETUSES ON DEVELOPING CHOROID PLEXUS. KARIOMETRIC STUDY International Journal of Morphology
Góes,M. J.; Azoubel,R..
The aim of the present study was to determine, by kariometric parameters, the influence of phenytoin, as teratogen, on the epithelial layer of choroid plexus in rats fetuses, using single daily dose ( 75 mg Kg-1 bd.wt.), during gestation GD9 to GD11, and lowest total dose used orally (225 mg Kg-1), to the best of our knowledge. In some of experimental studies in animals, the anticonvulsant drug phenytoin were administered in embriotoxic daily doses (100-1500 mg Kg-1 bd.wt.) increasing concentrations up to 20 times the human therapeutic plasma concentration. There is a significant lack of information regarding the embriotoxicity in pregnancy of these drug, phenytoin, in low doses. Phenytoin was administered in a single daily dose (75 mg Kg-1 bd.wt.) to...

Teratogenic Effect of Ethylene Glycol-Methyl Cellosolve Mixture in Rats.: I. Reproductive Damage International Journal of Morphology
Arteaga-Martínez,Manuel; García-Peláez,Isabel; Aguirre-Luna,Oswaldo M; Saavedra-Ontiveros,Dolores.
We investigated the reproductive damage and teratogenic effect of an ethylene glycol-methyl cellosolve mixture on gestating Wistar rats, which received a daily intraperitoneal dose of different concentration of the mixture on day 8 of gestation until day 20. In rats treated with the mixture the number of live fetuses decreased and reabsorptions increased with increasing concentrations of the mixture, as well as the number of abnormal fetuses. We conclude that the ethylene glycol-methyl cellosolve mixture possesses a higher teratogenic potential than each of its constituents separately, producing reproductive damage, external fetal abnormalities, growth delay, and increased fetal death.

Teratogenic Effect of Ethylene Glycol-Methyl Cellosolve Mixture in Rats: II. Craniofacial and Limb Abnormalities International Journal of Morphology
García-Peláez,Isabel; Aguirre-Luna,Oswaldo M; Saavedra-Ontiveros,Dolores; Arteaga-Martínez,Manuel.
We investigated the teratogenic effect of an ethylene glycol-methyl cellosolve mixture on gestating Wistar rats, that received a daily intraperitoneal dose of different concentration of the mixture on day 8 of gestation until day 20. Multivariate analysis and Post-Hoc Bonferroni tests were used and relative risk and attributable fraction were calculated. In rats treated with the mixture the number of live fetuses decreased and reabsorptions increased with increasing concentrations of the mixture, as well as the number of abnormal fetuses. Abnormalities consisted mainly in atypical craniofacial morphology, protruding tongue, edema, signs of growth delay and shorter limbs, their frequency and severity increased at higher concentrations of the mixture. We...
